“Phosphoantigens glue butyrophilin 3A1 and 2A1 to activate Vγ9Vδ2 T cells”
在癌症和感染中,患病的细胞通过一种“从内部向外”的信号传递过程被呈现给人类的Vγ9Vδ2 T细胞,这个过程是通过结构多样的磷酸抗原(pAg)分子被识别,这些分子被内源性的丁酸脂蛋白BTN3A1的细胞内结构域感知。
来自清华大学药学院张永辉及湖北大学郭瑞庭课题组合作展示了在人类和羊驼中,多种pAg分子充当“分子胶水”,促进BTN3A1和结构类似的丁酸脂蛋白BTN2A1的细胞内结构域的异源性结合。X射线晶体学研究可视化了BTN3A1与pAg的结合形成了一个用于直接与BTN2A1结合的复合界面,各种pAg分子分别位于界面的中心,并以不同的亲和力粘合着丁酸脂蛋白。使用基于结构的分子动力学模拟、19F-NMR研究、嵌合受体工程和直接测量细胞间结合力的分析,研究者揭示了pAg介导的BTN2A1结合如何以热力学有利的方式推动BTN3A1的细胞内波动向外扩散,从而使BTN3A1能够从BTN2A1外源结构域脱离,启动T细胞受体介导的γδ T细胞激活。从实际角度来看,研究者利用分子胶水模型进行了免疫治疗设计,展示了开发人类γδ T细胞功能的小分子激活剂和抑制剂的化学原理。
